Brassica campestris ssp. has been cultivated as oil and vegetable crop. However, it belongs to one of the dicot plant species recalcitrant to Agrobacterium infection for plant transformation and regeneration. We demonstrate that Agrobacterium-mediated transformation via vacuum infiltration poorly works for B. campestris var. narinosa seedlings in histochemical detection of GUS reporter gene expression. Either pre-germinated or germinating seeds were not susceptible to Agrobacterium infection. But, GUS gene expression was successfully achieved when 0.4% sodium hypochlorite solution, as an antimicrobial reagent, was treated for 2 min to two-days-old germinating seeds prior to agroinfiltration. Transformation efficiency was quantitated by using ELISA with another set of transgenic seedlings.
