Selection of Black Soybean Line with Absence of Kunitz Trypsin Inhibitor Protein

Soybean kunitz trypsin inhibitor protein is responsible for the inferior nutritional quality of unheated or incompletely heated soybean meal. The objective of this research was to select black seed coated soybean lines with absence of kunitz trypsin inhibitor protein. Black soybean genotype “20M183” (TiTi, kunitz trypsin inhibitor protein present, green cotyledon) was crossed with genotype “C242” (titi, kunitz trypsin inhibitor protein absent, yellow seed coat and cotyledon). Presence and absence of kunitz trypsin inhibitor protein was tested by SDS electrophoresis. Two F₂ plants with black seed coat, green cotyledon and absence of kunitz trypsin inhibitor were selected from the F₂ population. Each 10 F₃ plant from these F₂ plant selected was advanced. Seed coat color, cotyledon color and kunitz trypsin inhibitor protein were also tested after harvesting. Several F₄ seeds with black seed coat, green cotyledon and absence of kunitz trypsin inhibitor were planted to F₄ population (F₅ seed). Plant height, seed coat color, cotyledon color, seed weight, and kunitz trypsin inhibitor protein were estimated on the F₅ plants. Several F₅ plants with black seed coat, green cotyledon, 30 cm plant height, medium seed weight (20 g/100 seeds), and absence of kunitz trypsin inhibitor protein were selected. These plants will be used to improve the soybean cultivars with black seed coat, green cotyledon and absence of kunitz trypsin inhibitor protein.
