중국산 햄스터 난소세포로부터 활성형 Angiopoietin-1 생산에 관한 배양액 pH의 효과

Effect of Culture pH on Active Angiopoietin-1 Production from Chinese Hamster Ovary Cells

The effect of culture pH on cell growth and active Angiopoietin-1 production from the recombinant Chinese hamster ovary(CHO) cells was investigated. The cells in the bioreactor were incubated at different pH conditions(6.8, 7.0, 7.2 and 7.4). Specific growth rate decreased at lower culture pH while cell viability remained constant for a longer culture time. Consistently, glucose consumption and lactate production were also reduced at lower culture pH. In contrast to cell growth, expression level of active Angiopoietin-1 dramatically increased by lowering pH. In addition, Western blotting experiment at non-reduced and reduced condition showed that production of active Angiopoietin-1 forms significantly increased at lower pH. These results suggest that lowering culture pH may significantly improve production of biologically active Angiopoietin-1 from CHO cells.
