Ascorbic acid is a natural confound that is commonly biosynthesized in many animals except primates, guinea pigs, and some birds and possess an unusually high antioxidant activity. The aim of the present study was to investigate whether ascorbic acid could have anti-inflammtory effect in cells. In order to know whether the ascorbic acid would have the suppressive effect on the inflammatory mediators or cytokines gene expression. The levels of interleukin—6QL-6), tumor necrosis factor(TNF-a), cyclooxygenase-2(COX-2), RANTES, and inducible nitric oxide synthase(iNOS) expression were examined in lipopolysaccaride(LPS) - stimulated RAW 264.7 cells in the presence or absence of ascorbic acid treatment. LPS increased these genes expression in RAW264.7 cells. It was found that this increase was suppressed by ascorbic acid pre-treatment in LPS-stimulated RAW 264.7 cells. Ascorbic acid also suppressed reactive oxygen species(ROS) or reactive nitrogen species(RNS) production in LPS-stimulated RAW 264.7 cells. Therefore, these observations suggest that ascorbic acid can function as a regulator on the gene expression level.
