Analysis of a Before-After/Treatment-Control Experiment: A Nonparametric Conditional Two-Step Comparison on the Change Rate

사전-사후/처리-대조 실험으로부터의 자료에 대하여 많이 사용되는 분석들로 다음의 두 가지를 들 수 있다: (1) 사후 측정값과 사전 측정값 간의 차이를 구한 다음에, 처리 집단에서의 이러한 차이값들의 평균과 대조 집단에서의 이러한 차이값들의 평균을 비교하는 2표본 t-검정; (2) 사후 측정값을 반응변수, 사전 측정값을 공변량, 그리고 처리와 대조를 집단 설명변수의 2개의 수준들로 지정하는 공분산 분석. 그런데, 이러한 분석들이 유의한 결과를 내지 못하는 경우에도 처리와 대조 사이에는 어느 정도의 차이가 있을 수 있다. 본 연구는 이러한 차이를 탐지하는 것을 가능하게 하는, 사전-사후/처리-대조 실험의 분석방법으로 조건부 2단계 비교 절차를 제시한다. 사후 측정값과 사전 측정값 간의 단순한 차이보다는 이 차이를 사전 측정값으로 나눔으로써 얻어지는 변화율이 해석하기 쉬운 의미를 제공하기 때문에, 본 연구에서는 변화율을 분석대상으로 제시하였고, 변화율이 정규분포를 따른다고 보기 어려우므로, 변화율에 대한 평가 및 비교에서 비모수적 통계검정 방법들을 사용하였다. 과민성 대장 증후군 증상의 완화를 목적으로 하여 개발된 유산균 음료의 임상실험 자료를 이용하여 이 분석절차를 예시하였다.

For the analysis of a before-after/treatment-control experiment, we propose a nonparametric conditional two-step comparison procedure, suggesting the use of the change rate as a measure of change. The change rate is defined as “(A-B)/B”, where A denotes the value of a variable at the “after” time point and B denotes the value at the “before” time point. This change rate may not follow a normal distribution. Thus, there arises a need to use nonparameric techniques. At the first step of the proposed comparison procedure, in each of the treatment and control groups, the median of change rates is tested using the Wilcoxon signed-rank test for its significance, and these test results are compared between the two groups. The procedure proceeds to the second step only if the both test results are significant. At the second step of this comparison procedure, the two median change rates from the both groups are statistically compared using the Mann-Whitney rank-sum test. This procedure is illustrated using real data from an experiment to evaluate the effects of a newly-developed yogurt product on the symptoms of irritable bowel syndrome.
