We investigated the possible role of transient receptor potential vanilloid 1 (TRPV1) in melanogenesis in B16 melanoma cells. Capsaicin (CAP), a TRPV1 agonist, increased production of melanin and nitric oxide (NO) in a dosedependent manner, which was inhibited by capsazepine (CPZ), a TRPV1 antagonist. The CAP-induced melanin synthesis was blocked by L-NAME and cPTIO, an inhibitor of NO synthase and a NO scavenger, respectively. CAP increased intracellular Ca²⁺ concentration and BAPTA/AM, an intracellular Ca²⁺ chelator, inhibited the CAP-induced production of NO and melanin. These results suggest that TRPV1 may stimulate melanogenesis through intracellular Ca²⁺-mediated production of NO.
