The aim of this study is to quantify caffeic acid derivatives in Echinacea purpurea which has been used to as an immunomodulatory agent in various inflammatory and viral diseases. The root of Echinacea purpurea was extracted with methanol or 70%(v/v) methanol at different extraction conditions. The analysis was executed on an Eclipse plus-C18 column (4.6 mm × 150 mm, 5 μm) at 30 ℃ by gradient elution with a mobile phase of 0.085% (v/v) phosphoric acid in water and acetonitrile. The flow rate was 0.75 mL/min and detection was applied at 330 nm. Chromatographic separations of caffeic acid derivatives such as caftaric acid, caffeic acid and cichoric acid were exhibited and the correlation coefficients of caffeic acid derivatives in the test ranges were 0.9990~1.0000. The limits of detection and quantification values of the caftaric acid, caffeic acid and cichoric acid were 0.0068~0.0592 and 0.0.0227~0.1974 μg/mL, respectively. Quantitative analysis showed that caftaric acid, caffeic acid and cichoric acid were 6.751~7.440 mg/g, 0.428~0.451 mg/g and 10.875~12.446 mg/g in Echinacea purpurea by extraction with 70% methanol at room temperature, 40 ℃ and 40 ℃ with ultrasound treatment for 2 h, respectively.
