편백나무(Chamaecyparis obtusa) 추출물의 항산화, 미백효과에 관한 연구

본 연구는 화장품 소재로서 편백나무(Chamaecyparis obtusa)의 부위별 이용 가능성을 알아 보기 위하여 잎·가지(Leaf), 수피(Bark), 목재(Wood), 뿌리(Root)로 분리하여 99.9 % 에탄올에 추출한 시료로 항산화, 미백효능을 확인하였다. 이를 위해 다음과 같은 연구를 진행하였다. 항산화 평가는 1 1-diphenyl-2-picrylhydrazyl (DPPH) 라디칼 소거능, 2 2′-azino-bis (3-ethylbenzothiazoline--sulfonic acid) diammonium salt (ABTS + ) 라디칼 소거능으로 확인하였다. 미백 효과를 평가하기 위해서 mushroom tyrosinase 저해 활성 평가, MTT assay를 통한 편백나무 추출물의 세포독성평가를 진행 하였으며 cellular tyrosinase 저해율 측정과 melanin 생합성량을 확인하였다. 항산화 활성 결과 편백나무 부위 중 수피(Bark) 추출물이 가장 효능이 우수하였으며 B16F10 melanoma cell내 편백나무 수피(Bark) 는 tyrosinase 활성 억제 및 멜라닌 생합성 억제 효과를 나타내었다. 따라서 편백나무 수피 에탄올 추출 물의 안전하고 우수한 미백 기능성 화장품의 천연물질 소재로서의 개발 가능성을 확인하였다.
This study was conducted as follows in order to find out the possibility of using cypress tree (Chamaecyparis obtusa) by part as a cosmetic material. Chamaecyparis obtusa was separated leaves, bark, wood, and root, and extracted with 99.9% ethanol, and the antioxidant and whitening effects of the sample were confirmed. For this, the following studies were conducted. Antioxidant evaluation was confirmed by 1 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, 2 2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS + ) radical scavenging activity. To evaluate the whitening effect, mushroom tyrosinase inhibitory activity evaluation and cytotoxicity evaluation of Chamaecyparis obtusa extract through MTT assay were conducted, and cellular tyrosinase inhibition rate was measured and melanin contents was confirmed. As a result of antioxidant activity, Bark extract among Chamaecyparis obtusa parts showed the best efficacy, and bark in B16F10 melanoma cell showed tyrosinase activity inhibition and melanin contents inhibitory effects. Therefore, it was confirmed that the ethanol extract of Chamaecyparis obtusa was developed as a natural material for safe and excellent whitening functional cosmetics.
