Comparison of Antioxidant, Antimicrobial, and Anti-inflammatory Effects of Ethanol Extract from Neem(Azadirachta indica) and Its Fractions

In this study, the antioxidant, antimicrobial, and anti-inflammatory activities of a 70% ethanol extract of neem (Azadirachta indica) leafves and their fractions were evaluated to confirm the applicability as a functional cosmetic material. It was found that the total polyphenol content was higher in butanol (181.77±3.25 mg/g) and ethyl acetate fraction (180.27±1.53 mg/g) as compared to ethanol extract (139.16±1.04 mg/g). The electron donating ability and ABTS radical scavenging ability increased in a concentration-dependent manner in all samples. The IC50 value of electron donating ability was lower (357.6-375.1 ㎍/㎖) in ethyl acetate and butanol fractions compared to the than in ethanol extract (384.5 ㎍/㎖), and the ABTS radical scavenging activity was higher in the butanol fraction (66.8%) than the ethanol extract (64.3%) at the concentration of 1,000 ㎍/㎖. The antimicrobial activity of S. epidermidis and S. aureus were high in the ethyl acetate fraction, 8.5-11.5 mm and 8.5-15.0 mm, respectively, and the antibacterial activity against E. coli was high in the butanol fraction (8.5-14.0 mm). However, the antimicrobial activity against P. acne did not show a significant difference between the extract and the fraction. Cytotoxicity for RAW 264.7 cells was not observed up to the concentration of 100 ㎍/㎖ in both the extracts or fractions. The inhibition rate of NO production induced by LPS stimulation was highest in the ethyl acetate fraction (84.3%) than in the LPS alone treatment group, and about 2.3 times that of the ethanol extract (36.9%). Therefore, it was confirmed that the ethyl acetate fraction of neem leaves had a significant effect on the total polyphenol content, antioxidant, antibacterial activity, and inhibition of NO. In the future, it is considered necessary to study the anti-inflammatory mechanism of the ethyl acetate fraction.
