Antioxidant Effects of n-butanol Fraction of Coix Seed

To evaluate the antioxidant activity of coix seed (Coix lacryma-jobi L. var. mayuen Stapf), w e prepared the methanolic extract of coix seed and, subsequently, solvent polarity based frac tionation such as n-hexane, ethyl acetate, and n-butanol fraction, and then we determined th e content of the total polyphenols. The n-butanol fraction of coix seed (CLBu) displayed a hi gh level of polyphenols. And then we examined whether CLBu scavenges DPPH and ABTS fre e radicals, and possesses reducing power. CLBu demonstrated potent DPPH and ABTS free ra dical scavenging effects and a high level of reducing power. In addition, we investigated whe ther CLBu could inhibit the nitric oxide (NO) production in lipopolysaccharide (LPS)-activated RAW264.7 macrophages. The simultaneous cell viabilities showed CLBu protected the cell da mages from a high level of LPS-induced NO in RAW264.7 cells. These results suggested that CLBu may be a good source of antioxidant functional foods.
