Effect of High-Fat and High-Sucrose Diets on Secretion of Liver-Extracellular Vesicles in Mice

Extracellular vesicles (EVs) have been shown to provide a means of intercellular communication as well as contributing factors in the development of several diseases by EVs-mediated transfer of proteins, mRNAs, and microRNAs. In this study, we investigated the effects of high-fat (HFD) or high-fat/sucrose (HFD+SC) diets on EVs secretion in the liver to understand the underlying mechanisms associated with obesity-related metabolic diseases. Normal, HFD, and HFD+SC diets were fed to the mice for 8 weeks to compare differences based on dietary patterns, and then we harvested the EVs from each liver tissue. We found that the HFD and HFD+SC groups exhibited larger lipid droplets in both the liver and adipose tissues than the normal control group. However, the contents of lipid droplets in the liver were higher in the HFD+SC group than in the HFD group. In addition, the HFD+SC group showed a significant increase in EVs secretion from the liver than the HFD group. Therefore, we suggest that high-sugar intake not only accelerated the accumulation of lipid droplets in the liver but also hindered the secretion of extracellular vesicles. These results contribute to the elucidation of the mechanisms of EVs function in relation to obesity-related nonalcoholic fatty liver disease, and the metabolic relationship between tissues.
