Acute Inhalation Toxicity of Butanedioic Acid: A Study in Rats

Butanedioic acid is a water-soluble, colorless to white crystalline powder with no odor and acidic taste. It finds applications in cosmetics, medicine, and industry. Although oral toxicity data is available, inhalation toxicity data remains insufficient for comprehensive hazard assessment. To address this gap, we conducted an acute inhalation toxicity test according to the Organisation for Economic Co-operation and Development guidelines for the testing of chemicals (OECD TG) 436. We used a nose-only inhalation chamber system and a dust generator to expose the rats to test substances for 4 h. The actual (analytical) concentration was 4.878 ± 0.228 mg/L. Clinical signs, mean body weight changes, and gross necropsy results were recorded. No acute toxicity, including death, unusual clinical signs, gross necropsy findings, or abnormal mean body weight changes were observed. Based on these results, we suggest that butanedioic acid does not exhibit acute toxic effects at a concentration of 5 mg/L.
