Objectives The purpose of this study was to develop the highly effective and stand-ardized pharmacopuncture suitable for Chuna by extraction and purification process. Methods The electron donating ability of herbal medicines frequently used for joint and spine diseases was compared. Based on this comparison, Forsythiae Fructus and Lonicera japonica were selected for formulation and material name was JH002. The antioxidant activity of JH002 measured by 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), ferric reducing antioxidant power (FRAP), total flavonoids (TF), and total phenolic compounds (TP) assays. For standardization, index compounds were identified through monitoring, and method validation was conducted. Linearity, specificity, recovery, limit of quantification (LOQ), limit of detection (LOD), accuracy, and precision were measured to verify the validity of the index compounds. Results JH002 was highly active as a natural extract, and the selected forsythoside A from Forsythiae fructose and chlorogenic acid from Lonicera japonica were comparable as index compounds. Conclusions This study contributes not only to the development of JH002 but also to the standardization and scientific advancement of herbal medicine.
