This study investigated the in vitro anti-aging effects of tea flower polysaccharides obtained by ultrasound-assisted extraction (UAE-GTFP). Elastase and hyaluronidase inhibitory activities were also determined. UAE-GTFP showed no significant cytotoxicity at concentrations of 10-100 μg/mL, maintaining 95.9% cell viability even at 100 μg/mL. UAE-GTFP significantly reduced H₂O₂-induced SA-β-gal-positive cells in a concentration-dependent manner. Furthermore, UAE-GTFP restored the UVB-induced reductions in type I procollagen, elastin, and hyaluronic acid production while suppressing MMP-1 production. At 100 μg/mL, type I procollagen and elastin production exceeded the untreated control levels, whereas hyaluronic acid production was significantly higher than the control at concentrations of 50 μg/mL or greater. UAE-GTFP also inhibited elastase and hyaluronidase activities in a concentration-dependent manner, reaching 76.8% and 87.0% inhibition, respectively, at 100 μg/mL. These findings indicate that UAE-GTFP protects skin fibroblasts against oxidative stress and UVB-induced damage by promoting extracellular matrix synthesis and inhibiting its enzymatic degradation. Therefore, UAE-GTFP has considerable potential as a natural functional ingredient for improving skin elasticity and hydration and preventing skin photoaging.
