Safety Analysis of Solutions for Washing of Contact Lenses

Acanthamoeba is a free-living protozoan pathogen which causes blinding amoebic keratitis (AK) to contact-lens users. In this study, using commercialized contact lens-clearing solutions, their cytotoxicity were analyzed against Acanthamoeba. Moreover, their effects to Acanthamoeba were compared with cytotoxicity to a human corneal epithelial cell (HCEC) as a target cell. Only 10% of the lens solution of Amo Total Care 1 didn"t significantly affect the morphology of A. castellanii. As the percent of the lens solution was increased, it decreased the number of A. castellanii and changed shapes of the amoebae. 10% and 50% of Opti-free RepleniSH decreased the number of A. castellanii more than Amo Total Care 1. 0 to 100 ul of lens solutions had little cytotoxicity to HCEC. A. castellanii was severely affected about 40% of cytotoxicity of Amo Total Care 1. On the other hand, Opti-free RepleniSH had about 38% to 58% cytotoxicity to A. castellanii Our results showed that two contact-lens clearing solutions didn"t have very strong effects to A. castellanii trophozoites. Therefore, it suggests that other compositions and compounds of stronger cytotoxicity should be developed for disinfections to A. castellanii.
