The Author measured plasma prolactin level with the method of RIA upon 14 normal male volunteers to study the effect of benztropine on the prolactirt response to haloperidol. With the interval of 7 days, plasma prolactin levels were measured on 15,30,45,60,90, and 120 minutes after administration with and without benztropine, and the results were compared with baseline plasma prolactin level already measured. The results were as below; 1. The mean baseline plasma prolactin level in normal volunteers was 10. 75 ±3. 28ng/ml with individual variance. 2. The peak plasma level (21. 75ng/ml) appeared on 60 minutes after administration of haloperidol (0. 5mg). 3. With administration of both haloperidol (0. 5mg) and benztropine (0. 5mg), the mean plasma prolactin level was increasing steadily. 4. Comparing these tw o groups, increment of the plasma prolactin level was higher during the earlier phase in the group of haloperidol alone, and this relationship was reversed between 60 and 90 minutes after administration. 5. The plasma level in the group of haloperidol alone on 15 minutes was significantly higher ( p < 0 .02), and significantly lower ( p < 0 .02) on 120 minutes than that in the group of combined administration. With these results, clinical implication was discussed briefly
