腦電氣衝撃이 흰쥐 大腦組織内의 核酸 및 蛋白質 含量에 미치는 影響

EFFECTS OF CEREBRAL ELECTROCONVULSIVE SHOCK ON THE CONTENTS OF NUCLEIC ACIDS AND PROTEIN IN THE CEREBRAL TISSUE OF ALBINO RAT

The author performed the following experiment to study the effects of electroconvulsive shock on the contents of nucleic acids and protein, and their respective biosynthetic activities in the cerebral tissue of albino rats. A single electroconvulsive shock (ECS, AC, 60 Hz, 110 volt, 30mA, 0 .02minute) was applied to Sprague-Dawley albino rat through bitemporal electrodes with Sedac Stimulator, Model SOS No. 1722 (Reuben Reiter Company). As a function of time following single ECS, the contents of nucleic acids, protein and TCA-soluble aminoacid in the cerebral tissue were analysed. At the same time, protein and R N A biosynthetic activities were determined. To clarify the mode of protein and nucleic acid biosynthetic alteration, polysome/monosome ratio and alkaline RNase activity were simultaneously assayed, The results are as follows ; 1. The content of DNA shows no significant change, while those of protein, RNA and TCAsoluble amino acids decrease and resume within 24 hours, respectively. 2. The biosynthetic activity of protein as observed by leucine-3-C14 incorporation decreases immediately and recovers w ith in 30 minutes after ECS, while that of RNA, by* orotic acid- 6-C14 incorporation shows delayed recovery and even increased activity. 3. Polysome/monosome ratio, as determined by sucrose density gradient ultracentrifuge method, decreases after ECS, but w ith in 1 hour, returns to the control level and shows “rebound” activity after an hour. The free alkaline RNase activity increases at 30 minutes after EES. But it returns to control level at 1 hour after ECS and shows decreasing tendency after 2 hours following ECS below the control level.
