In this experiment, the author observed the change in the plasma concentration of ACTH and corticosterone, reflecting some of H P A axis, after administration of Imipramine into rats. The purpose of this experiment was to know mechanism of action of Imipramine via neuroendocrinological changes in H P A axis after administration of Imipramine, a tricyclic antidepressant. Rats were divided into 2 groups, Imipramine and control group, then each group divided into 4 groups, 1, 2, 4 and 8 week groups. Plasma of the 1st, 2nd, 4th, 8th week after the administration was obtained from each group and the concentration of A C T H and corticosterone was measured by radioimmunoassay. The results were as follows ; 1) There was statistically nosignificant difference between Imipramine and control group each week in the concentration of plasma ACTH . 2) There was statistically nosignificant difference between Imipramine and control group each week in the concentration of plasma corticosterone. 3) There was statistically no significant change from 1 to 8 weeks in the concentration o f plasma A C T H and corticosterone. This finding showed that there was no changes of pituitary-adrenal axis after administration of Imipramine. But this finding could not exclude the possibility that there was change above hypothalamus level, thus further studies for this aspccts will be necessary
