Desipramine 장기투여가 백서 대뇌피질의 Adenylate Cyclase 의 활성도와 GABAb 및 Noradrenaline 수용체후 구조의 Coupling system 에 미치는 영향

Effect of Chronic Desipramine Administration on the Forskalin Stimulated Adenylate Cyclase Activity and the Coupling of GABAb and Norcpinq)hrine-cAMP Generating System

Long-term treatment of rats with the antidepressant desipramine led to an increase in the potentiation of isoproterenol-induced cAMP accumulation by baclofen in cortical slice. However, the identical treatment with desipramine did not increase in GABAb binding. These results suggest that the mechanism responsible for this potentiation effect is not simply to direct GABAb receptor stimulation. Forskolin-stimulated adenylate cyclase activity increased in cortical slice from rats which received long-term desipramine treatment, while other previous studies have presented inconsistent results. The author assumes that the change of post-5-adrenoceptor component may be more relevant than receptor down-regulation to the therapeutic action of tricyclic antidepressants. However verification of this hypothesis will need further investigation and more elabolate experiments.
