The authors conducted an open-trial clinical study to evaluate the clinical efficacy of toloxatone, a reversible inhibitor of MAO-A. The study group were 8 6 patients with either DSMIII- R diagnosis of major depression or dysthymia, who visited the Department of Psychiatry College of Medicine, Yeungnam University during the period from January to July 1992 and each patient received toloxatone(600mg/day) for 28 days. The clinical effect of toloxatone were evaluated using the Hamilton Rating Scale for Depression, weekly by two psychiatrists. The results were as follows : 1) There was statistically significant improvement on the Hamilton Rating Scale for Depression after 7 days treatment(p<0.01). 2) Side effects were excitement and insomnia, gastrointestinal effects, drowsiness, dizziness, anticholinergic effects and facial edema in a few patient, but were tolerable. 3) In all subjects, hypertensive reaction was not observed without food restriction and blood pressure did not show any differences
