Ultraviolet B (UVB), which ranges from 280 to 320 nm, induces DNA damage, including the formation of pyrimidine dimer such as cyclobutane pyrimidine dimers (CPDs). CPDs are identified by cleavage through repair enzymes such as T4 PDG. Shift of plasmid with DNA breakage on agarose gel could be used for identification of DNA damage by UVB. However, without double strand break on plasmid, it is difficult to identify the extents of CPDs generation by UVB through treatment of T4 PDG alone. In this study, we examined whether treatment with T4 PDG to UVB-irradiated plasmid followed by NaOH addition could show the DNA damage clearly.
