Inhibitory Effect of Machilus thunbergii Extract on Melanoma Cell Proliferation

This study aimed to evaluate the anticancer and anti-inflammatory effects of Machilus thunbergii extract (MTE) on the B16F10 melanoma cell line and to elucidate its underlying mechanisms. Melanoma, which arises from the transformation of melanocytes, necessitates effective treatment due to its rapid growth and metastasis. We treated B16F10 cells with MTE and assessed changes in the expression of cancerrelated genes using Western blot and qRT-PCR. Cell viability was analyzed across varying concentrations and exposure times using the WST assay. The wound healing assay revealed that MTE effectively inhibited the recovery of melanoma cells. Additionally, changes in the expression of cancerrelated genes were examined using qRT-PCR. The results showed that MTE increased the expression of cancer genes such as FGF, CDK4, and C-Myc, while significantly suppressing the proliferation and migration of cancer cells. Notably, higher concentrations of MTE led to a significant reduction in cell viability, and qRT-PCR analysis revealed decreased expression of inflammatory genes TGF-beta, TNF-alpha, and F4/80. These findings suggest that MTE has the potential to act as a natural anticancer agent, inhibiting melanoma cell proliferation and exhibiting anti-inflammatory effects, warranting further research for its potential application in melanoma treatment.
