Integrative Multi-Omics Analysis Reveals CDKN2A as a Potential Diagnostic and Prognostic Biomarker in Cervical Cancer

Cervical cancer remains a major malignancy with substantial global morbidity and mortality, and identifying molecular determinants beyond HPV infection is essential. In this study, we conducted an integrative multi-omics analysis to discover key genes involved in cell-cycle dysregulation and to assess their diagnostic and prognostic relevance. KEGG analysis of publicly available transcriptomic datasets showed that the cell cycle pathway was the most significantly upregulated in cervical cancer. Cross-analysis of independent GEO datasets highlighted CDKN2A as a consistently elevated cell-cycle-related gene, and additional transcriptomic and proteomic data further confirmed its increase at both mRNA and protein levels. In the TCGA cohort, high CDKN2A expression was associated with significantly poorer overall survival. Single-cell RNA sequencing also demonstrated that CDKN2A expression was predominantly localized to epithelial cells, the major cellular component of cervical cancer. Collectively, these findings identify CDKN2A as a central marker of cell-cycle dysregulation in cervical cancer and support its potential clinical utility as a diagnostic and prognostic biomarker.
